It depends on how hard you are pushing the 45 Microblock. If you have ample headroom and not turning all the way up, the 25 watt might be enough. But if you are pushing the 45 Microblock to its limits, the 25 watt won't provide any more power.

Yes, the Quilter Superblock is designed to be mostly used for line out to the mix board.

Is the preamp new-in-box?

Based on my experience, mine seemed to be new-in-box. It was immaculately packed in its Quilter box. I am quite happy with it.
